The present invention comprises rugged, inexpensive, reliable, and sensitive laboratory assays of antibody-based viral neutralization activity and antibody-based viral adherence inhibition activity. The assays use inactivated, fluorescently-labeled virus, allowing the tests to be performed without extensive safety precautions. The interaction of the labeled virus with target cells is monitored using flow cytometric methods. A preferred embodiment uses simple and inexpensive flow cytometry methodologies and equipment, such as bead array readers used as simplified flow cytometers. The assays are rapid, taking no longer than a few hours and are readily conducted by a trained technician. The assays are sensitive because they use labeled viruses at low concentrations and determine neutralizing and blocking capacity of sera and antibody at low concentrations. The methods are appropriate for high-throughput screening of large panels of samples.

La présente invention concerne des dosages de laboratoire bien construits, bon marché, fiables et sensibles d'une activité de neutralisation virale à base d'anticorps et d'une activité d'inhibition d'adhésion virale à base d'anticorps. Ces dosages utilisent des virus inactivés marqués par fluorescence, ce qui permet d'effectuer les essais sans grandes précautions de sécurité. L'interaction d'un virus marqué avec des cellules cibles est suivie par des méthodes de cytométrie en flux. Un mode de réalisation préféré repose sur des méthodologies et un équipement de cytométrie en flux simples et bon marché tels que des lecteurs de réseaux de billes utilisés comme cytomètres en flux simplifiés. Les dosages sont rapides, ne prenant pas plus de quelques heures, et sont facilement réalisés par un technicien formé. Les dosages sont sensibles, car ils utilisent des virus marqués à de faibles concentrations et déterminent la capacité de neutralisation et de blocage de sérums et d'anticorps à de faibles concentrations. Les procédés ci-décrits sont adaptés à un criblage à haut débit de grands panels d'échantillons.
