(EN)Methods and compositions for cloning a donor DNA molecule into an acceptor vector at a predetermined location are described. The methods are based on homologous recombination mediated by in vitro treatment of the donor DNA and the acceptor vector with an enzyme cocktail containing an exonuclease and a single-stranded DNA binding protein.
(FR)La présente invention concerne des procédés et des compositions pour cloner une molécule d’ADN donneuse dans un vecteur accepteur à un emplacement prédéterminé. Les procédés sont basés sur une recombinaison homologue dont la médiation est assurée par un traitement in vitro de l’ADN donneur et du vecteur accepteur avec un cocktail d’enzymes contenant une exonucléase et une protéine de liaison d’ADN monocaténaire.
