(EN)The invention provides antibacterial compounds comprising an oligonucleotide having a sequence complementary to a translation initiation region of an mRNA encoding a mycolyl transferase of Mycobacterium tuberculosis selected from protein 30, 32A and 32B, and further having 5 ' and 3 ' palindromic hairpin- forming sequences, said compound being covalently linked to a protein synthesis inhibiting antibiotic via a linker. The invention further provides pharmaceutical formulations of such compounds and methods of use thereof for treating tuberculosis. Other diseases may similarly be treated by tethering and antibiotic which targets a ribosomal protein to an antisense oligonucleotide complementary to an mRNA involved in the disease.
(FR)La présente invention concerne des composés antibactériens comprenant un oligonucléotide ayant une séquence complémentaire d'une région d'initiation de la traduction d'un ARNm codant pour une mycolyl transférase du bacille de Koch choisi parmi les protéines 30, 32A et 32B, et ayant en outre des séquences formant une boucle en épingle à cheveux 5' et 3' palindromique, ledit composé étant lié de manière covalente à un antibiotique inhibant la synthèse des protéines par le biais d'une séquence de liaison. L'invention concerne en outre des formulations pharmaceutiques de tels composés et leurs procédés d'utilisation dans le traitement de la tuberculose. D'autres maladies peuvent être traitées de manière similaire en insérant un antibiotique qui cible une protéine ribosomale sur un oligonucléotide antisens complémentaire d'un ARNm impliqué dans la maladie.
