(EN)Disclosed is a method for producing a cDNA library having a reduced content of a cDNA clone derived from a highly expressed gene with high efficiency. In a method for producing a cDNA library by using a double-stranded DNA primer containing an oligo-dT and also using mRNA as a template, the content of a cDNA clone derived from a highly expressed gene in the cDNA library can be reduced by allowing a probe to coexist, wherein the probe has such a property that the probe can bind to mRNA of a highly expressed target gene to inhibit the occurrence of a cDNA extension reaction with a reverse transcriptase.
(FR)L’invention concerne un procédé de fabrication d’une bibliothèque d’ADNc ayant une teneur réduite en un clone d’ADNc dérivé d’un gène hautement exprimé avec une grande efficacité. Selon un procédé de fabrication d’une bibliothèque d’ADNc qui utilise une amorce d’ADN bicaténaire contenant un oligo-dT et qui utilise également un ARNm en tant que matrice, la teneur en un clone d’ADNc dérivé d’un gène hautement exprimé dans la bibliothèque d’ADNc peut être réduite en autorisant la coexistence d’une sonde, la sonde ayant une propriété telle que la sonde peut se lier à l’ARNm d’un gène cible hautement exprimé pour inhiber l’occurrence d’une réaction d’extension d’ADNc avec une transcriptase inverse.
(JA) 高発現遺伝子由来のcDNAクローンの含有率を低減させたcDNAライブラリーを効率的に作製する方法を提供すること。  オリゴdTを有する二本鎖DNAプライマーを用いてmRNAを鋳型にしてcDNAライブラリーを作製方法において、高発現している標的遺伝子のmRNAに結合して逆転写酵素によるcDNA伸長反応を阻止する特性のあるプローブを共存させることによって、cDNAライブラリー中の高発現遺伝子由来cDNAクローンの割合を低減する。
