Abstract

Disclosed is a novel lectin which can bind specifically to a fucose α1→6 sugar chain. Also disclosed is use of the lectin. Specifically disclosed is a fucose α1→6-specific lectin characterized by the following properties (1) to (3): (1) the lectin can be extracted from a basidiomycete or an ascomycete; (2) the lectin has a molecular weight of 4,000 to 40,000 as measured by an SDS electrophoresis method; and (3) the lectin has affinity for a fucose α1→6 sugar chain at a binding constant of 1.0 × 104 M-1 or more (at 25°C). The fucose α1→6-specific lectin can be used for detecting a fucose α1→6 sugar chain specifically, and is effective for the purification of a fucose α1→6 sugar chain or a non-fucose α1→6 sugar chain.

La présente invention concerne une nouvelle lectine qui peut se lier spécifiquement à une chaîne de sucre fucose α1→6. L'invention concerne également l'utilisation de la lectine. L'invention concerne spécifiquement une lectine spécifique du fucose α1→6 caractérisée par les propriétés (1) à (3) suivantes : (1) la lectine peut être extraite d'un basidiomycète ou d'un ascomycète; (2) la lectine a un poids moléculaire, allant de 4000 à 40000, mesuré par un procédé d'électrophorèse SDS; et (3) la lectine a une affinité pour une chaîne de sucre fucose α1→6 à une constante de liaison de 1,0 × 104 M-1 ou plus (à 25 °C). La lectine spécifique du fucose α1→6 peut être utilisée pour la détection spécifique d'une chaîne de sucre fucose α1→6, et est efficace pour la purification d'une chaîne fucose α1→6 ou d'une chaîne de sucre non-fucose α1→6.

【課題】フコースα1→6糖鎖に特異的に結合する新規レクチンとその用途を提供する。【解決手段】本発明のフコースα1→6特異的レクチンは、(1)担子菌又は子嚢菌から抽出され、(2)SDS電気泳動法による分子量が4,000~40,000であり、及び、(3)フコースα1→6糖鎖に対して結合定数1.0×10-1以上(25℃において)で示される親和性を有することを特徴とする。これを用いれば、フコースα1→6糖鎖を特異的に検出し、また、フコースα1→6糖鎖又は非フコースα1→6糖鎖の精製に有効である。
