In the production of a cell mass containing a pluripotent stem cell, a step of treating a somatic cell under nutrient-starved conditions while contacting the somatic cell with a nuclear reprogramming factor and/or a step of treating the somatic cell with a substance capable of arresting a cell cycle is involved.  It becomes possible to induce and grow a pluripotent stem cell at high frequency, and it also becomes possible to produce a pluripotent stem cell with high efficiency.  The nuclear reprogramming factor to be used may be any one selected from the group consisting of OCT4, SOX2, c-MYC, KLF4, NANOG and LIN28.

Cette invention concerne, dans le cadre de la production d’une masse cellulaire contenant une cellule souche pluripotente, une étape de traitement d’une cellule somatique dans des conditions appauvries en nutriments tandis que la cellule somatique est mise en contact avec un facteur de reprogrammation nucléaire et/ou une étape de traitement de la cellule somatique avec une substance capable de stopper un cycle cellulaire. Il devient alors possible d’induire et de faire croître une cellule souche pluripotente à une fréquence élevée, et il devient aussi possible de produire une cellule souche pluripotente avec une grande efficacité. Le facteur de reprogrammation nucléaire à utiliser peut être l’un quelconque des facteurs choisis dans le groupe constitué par OCT4, SOX2, c-MYC, KLF4, NANOG et LIN28.

 本発明によれば、多能性幹細胞を含有する細胞集団を製造する際において、核初期化因子と接触させた体細胞を栄養飢餓条件で処理する工程及び/又は細胞周期を停止させる薬剤で処理する工程を包含させる。これにより、高い頻度で多能性幹細胞を誘導、成育させることができ、多能性幹細胞を高効率で製造することができる。当該核初期化因子としては、例えば、OCT4、SOX2、c-MYC、KLF4、NANOG及びLIN28からなる群より選択される核初期化因子が使用できる。
