Aspects of the present invention are drawn to methods and compositions for sorting nucleic acid molecules into physically separate compartments according to the identity of a nucleotide base or sequence of bases at a specific location, resulting in the production of reduced complexity samples that find use in any number of downstream genetic analyses. Aspects of the methods of the invention include fragmenting a nucleic acid sample, e.g., with a restriction enzyme, ligating an adaptor (or adaptors), and sorting the fragments based on the identity of the nucleotide base(s) positioned adjacent to the fragmentation site (e.g., the restriction enzyme cut site/or recognition site). Each round of sorting produces binned samples having reduced complexity over the parent sample.

Selon des aspects, la présente invention concerne des méthodes et des compositions pour trier des molécules d'acides nucléiques dans des compartiments physiquement séparés en fonction de l'identité d'une base nucléotidique ou d'une séquence de bases à un emplacement spécifique, entraînant la production d'échantillons de complexité réduite qui peuvent être utilisés dans nombre d'analyses génétiques en aval. Selon des aspects, les méthodes de l'invention consistent à fragmenter un échantillon d'acides nucléiques, par ex., à l'aide d'une enzyme de restriction, à ligaturer un adaptateur (ou des adaptateurs), et à trier les fragments en fonction de l'identité de la/des base(s) nucléotidique(s) positionnée(s) de manière adjacente au site de fragmentation (par ex., le site de coupure/ou site de reconnaissance de l'enzyme de restriction). Chaque cycle de tri produit des échantillons regroupés présentant une complexité réduite par rapport à l'échantillon parent.
