Abstract
(EN) A method of preparing a population of in vitro cultured cells of alveolar epithelial type II (ATII) cell lineage derived from at least one embryonic stem cell is disclosed which comprises (a) culturing said at least one embryonic stem cell in vitro in a medium comprising Matrigel®, to produce differentiated cells without formation of an embryonic body, wherein at least some of the differentiated cells are of ATII cell phenotype; (b) identifying the differentiated cells of ATII cell phenotype by detecting expression of at least one biomarker of ATII cells; (c) isolating the differentiated cells having ATII cell phenotype; and (d) cloning the isolated cells to produce a population of cells having ATII cell phenotype. The resulting cells are preferably >99% pure ATII phenotype lineage and are potentially useful therapeutically for treating lung injury and disease.
(FR) L'invention concerne un procédé de préparation d'une population de cellules cultivées in vitro d'une lignée cellulaire épithéliale alvéolaire de type II (ATII) dérivée d'au moins une cellule souche embryonnaire, ledit procédé comprenant les étapes consistant à : (a) cultiver ladite ou lesdites cellules souches embryonnaires in vitro dans un milieu comprenant Matrigel® pour produire des cellules différenciées sans formation d'un corps embryonnaire, au moins certaines des cellules différenciées étant de phénotype cellulaire ATII; (b) identifier les cellules différenciées de phénotype cellulaire ATII en détectant l'expression d'au moins un biomarqueur des cellules ATII; (c) isoler les cellules différenciées ayant le phénotype cellulaire ATII; et (d) cloner les cellules isolées pour produire une population de cellules ayant le phénotype cellulaire ATII. Les cellules résultantes sont de préférence une lignée de phénotype ATII pure à plus de 99 % et sont potentiellement utiles du point de vue thérapeutique pour traiter les blessures et les maladies pulmonaires.
