Methods, processes and systems for procuring, isolating and cryopreserving at least one viable, multipotent maternal placental stem cell is provided. Viable maternal placental stem cells are also provided. The maternal placental stem cell of the invention expresses the cell surface marker CDl 17 and at least one of the cell surface markers selected from the group consisting of CD29, CD44, CD73, CD90, CD105, CD166, SSEA-3 and SSEA-4 and has low or no expression of at least one of the cell surface markers selected from the group consisting of CD34, CD45, CD133, TRA-1-60 and TRA-1-81. The methods and process comprise generally obtaining a piece of placental tissue from a whole placenta, disaggregating the placental tissue with mechanical separation or enzymatic digestion, collecting and concentrating placental cells comprising maternal placental cells with centrifugation and optionally cryopreserving the placental cells. Additional steps for selecting and culturing the maternal placental stem cells is provided.

L'invention concerne des méthodes, des procédés et des systèmes d'obtention, d'isolation et de cryoconservation d'au moins une cellule souche placentaire maternelle multipotente viable. Elle concerne également des cellules souches placentaires maternelles viables. La cellule souche placentaire maternelle de l'invention exprime le marqueur de surface cellulaire CD1 17 et au moins un marqueur de surface cellulaire choisi dans le groupe constitué par CD29, CD44, CD73, CD90, CD105, CD166, SSEA-3 et SSEA-4 et elle présente une expression faible ou nulle d'au moins un marqueur de surface cellulaire choisi dans le groupe constitué par CD34, CD45, CD133, TRA-1-60 et TRA-1-81. Les méthodes et les procédés comprennent de manière générale l'obtention d'un morceau de tissu placentaire à partir de placenta entier, la dissociation du tissu placentaire par séparation mécanique ou digestion enzymatique, la collecte et la concentration de cellules placentaires comprenant des cellules placentaires maternelles par centrifugation, et le cas échéant la cryoconservation des cellules placentaires. L'invention concerne d'autres étapes de sélection et de culture des cellules souches placentaires maternelles.
