Disclosed is a method for increasing a melting temperature (Tm) of double-stranded DNA. Also disclosed is a reagent for use in the method. More specifically, disclosed is an agent for increasing a melting temperature (Tm) of a DNA molecule having a G-G sequence composed of two contiguous guanine (G) residues and also having a G-G mismatch and at least one other mismatch, which comprises a naphthyridine carbamate dimer derivative represented by the general formula (1): (1) wherein R1 and R2 independently represent a hydrogen atom or a group such as a hydrocarbon group; R represents an alkylene group; X represents an oxygen or sulfur atom; and R3 represents a hydrogen atom, an alkyl group having 1 to 5 carbon atoms, or a group represented by the general formula (2): (2) wherein R1 and R2 independently represent a hydrogen atom or a group such as a hydrocarbon group; R represents an alkylene group; X represents an oxygen or sulfur atom; and L represents a linker group for binding two nitrogen atoms together. Also disclosed is a method using the agent.

La présente invention concerne un procédé destiné à augmenter une température de fusion (Tf) d'ADN double brin. La présente invention concerne également un réactif destiné à être utilisé dans ledit procédé. Plus spécifiquement, l'invention concerne un agent destiné à augmenter une température de fusion (Tf) d'une molécule d'ADN présentant une séquence G-G composée de deux résidus contigus de guanine (G), et présentant également un mésappariement G-G ainsi qu'au moins un autre mésappariement, qui comprend un dérivé de dimère de carbamate de naphthyridine représenté par la formule générale (1) : (1,) R1 et R2 représentant indépendamment un atome d'hydrogène ou un groupe tel qu'un groupe hydrocarbure ; R représentant un groupe alkylène ; X représentant un atome d'oxygène ou de soufre ; et R3 représentant un atome d'hydrogène, un groupe alkyle présentant 1 à 5 atomes de carbone, ou un groupe représenté par la formule générale (2) : (2), R1 et R2 représentant indépendamment un atome d'hydrogène ou un groupe tel qu'un groupe hydrocarbure ; R représentant un groupe alkylène ; X représentant un atome d'oxygène ou de soufre ; et L représentant un groupe lieur destiné à lier deux atomes d'azote.
