Die Erfindung betrifft ein Verfahren zum Auffinden geeigneter Parameter für die chromatographische Reinigung von Biomolekülen. Das Verfahren besteht aus Equilibrierungs-, Beladungs-, Wasch- und Elutionsschritten, wobei diese Schrittfolge im teilweisen Satzverfahren durchgeführt wird. Die in kleinen, vorzugsweise mehreren parallelen Ansätzen ermittelten Parameter geben Aufschluss auf die Chromatographiebedingungen, unter denen sich ein gegebenes Biomolekül optimal säulenchromatographisch, ggf. auch in größerem Maßstab, reinigen lässt.

The invention relates to a process for finding suitable parameters for the chromatographic purification of biomolecules. The process consists of equilibration, loading, washing and elution steps, wherein this sequence of steps is carried out in the partial batch process. The parameters determined in small, preferably a plurality, of parallel batches give information on the chromatography conditions under which a given biomolecule may be optimally purified by column chromatography, if appropriate even on a relatively large scale.

L'invention concerne un procédé permettant de trouver des paramètres appropriés pour la purification chromatographique de biomolécules. Ce procédé comprend des étapes d'équilibrage, de chargement, de lavage et d'élution, cette séquence d'étapes pouvant s'effectuer en partie par lots. Les paramètres déterminés dans de petites préparations, de préférence plusieurs en parallèle, donnent des indications sur les conditions chromatographiques qui permettent de purifier une biomolécule donnée de manière optimale par chromatographie sur colonne, y compris le cas échéant à grande échelle.
