The present invention relates to methods for sequencing a polynucleotide immobilized on an array having a plurality of specific regions each having a defined diameter size, including synthesizing a concatemer of a polynucleotide by rolling circle amplification, wherein the concatemer has a cross-sectional diameter greater than the diameter of a specific region, immobilizing the concatemer to the specific region to make an immobilized concatemer, and sequencing the immobilized concatemer.

La présente invention concerne des procédés permettant le séquençage d'un polynucléotide immobilisé sur un réseau comportant une pluralité de régions spécifiques, possédant chacune un diamètre défini. Les procédés consistent à synthétiser un concatémère d'un polynucléotide par amplification en cercle tournant, le concatémère présentant un diamètre en coupe transversale supérieur au diamètre d'une région spécifique, à immobiliser le concatémère sur la région spécifique afin d'en faire un concatémère immobilisé et à séquencer ledit concatémère immobilisé.
