[PROBLEMS] To provide a carrier for immunoglobulin affinity chromatography that simultaneously has high immunoglobulin binding capability and chemical stability and that can be produced at low cost. [MEANS FOR SOLVING PROBLEMS] An immunoglobulin binding protein wherein an amino acid substitution for not only maximizing the number of lysine residues on the protein surface of helix 3 and its periphery but also minimizing the number of lysine residues present on the protein surface of helix 1 and helix 2 as immunoglobulin binding regions and/or an amino acid substitution for eliminating of asparagine acid-proline sequence has been carried out, or a multimer thereof, is used as an affinity ligand of carrier for affinity chromatography.

Le problème à résoudre dans le cadre de cette invention consiste à trouver un véhicule pour la chromatographie à affinité sur immunoglobulines qui possède à la fois une haute capacité de liaison aux immunoglobulines et une stabilité chimique, et qui puisse être produit à faible coût. La solution proposée consiste à utiliser une protéine de liaison aux immunoglobulines, dans laquelle on effectue une substitution d'acides aminés non seulement pour maximiser le nombre de résidus de lysine présents sur la surface protéique de l'hélice 3 et sa périphérie, mais aussi pour réduire au minimum le nombre de résidus de lysine présents sur la surface protéique de l'hélice 1 et de l'hélice 2 en tant que régions de liaison aux immunoglobulines et/ou une substitution d'acides aminés visant à éliminer la séquence acide asparaginique–proline, ou d'utiliser un multimère de celle-ci, en tant que ligand d'affinité d'un véhicule pour la chromatographie à affinité.

【課題】 本発明は、高いイムノグロブリン結合能と化学的安定性を兼ね備え、かつ安価に製造可能なイムノグロブリンのアフィニティクロマトグラフィー用担体を提供することを課題とする。 【解決手段】 イムノグロブリン結合領域であるヘリックス1およびへリックス2のタンパク質表面に存在するリジン残基数を可能な限り減少させるとともに、へリックス3およびその周辺のタンパク質表面に可能な限りリジン残基数を増加させるアミノ酸置換および/またはアスパラギン酸-プロリンの配列を排除するアミノ酸置換を行ったイムノグロブリン結合タンパク質、もしくはその多量体を、アフィニティクロマトグラフィー用担体の親和性リガンドとして用いる。
