The invention relates to a recombinant cell-based system in which the expression of the Lassa Virus receptor in one cell and the expression of the GP-C protein from Lassa Virus in another cell is used to have cells fused and thereby to mimic cellular entry of Lassa Virus. The system of the invention further uses reporter genes in one cell population and a transactivator protein in the other cell population to quantify the occurring cell fusion. The system is applied for screening anti-fusion-acting compounds that are thus identified as having anti -Lassa Virus activity. Such compounds are applicable in the treatment of Lassa Virus- induced disease.

La présente invention concerne un système à base de cellules recombinées dans lequel l'expression du récepteur du virus de Lassa dans une cellule et l'expression de la protéine GP-C provenant du virus de Lassa dans une autre cellule servent à fusionner les cellules et ainsi à imiter l'entrée cellulaire du virus de Lassa. Le système de l'invention utilise aussi des gènes rapporteurs dans une population de cellules et une protéine transactivatrice dans l'autre population de cellules afin de quantifier la fusion cellulaire qui se produit. Le système s'applique lors du criblage de composés agissant contre la fusion qui sont ainsi identifiés comme présentant une activité antivirale à l'encontre du virus de Lassa. De tels composés peuvent être utilisés lors du traitement de maladies induites par le virus de Lassa.
