Abstract:
(EN) Well-defined, xeno-free culture media which comprise a TGF-beta isoform or the chimera formed between IL6 and the soluble IL6 receptor (IL6RIL6), which are capable of mainataining stem cells, and particularly, human embryonic stem cells, in an undifferentiated state are provided. Also provided are cell cultures comprising the culture media and the stem cells and methods of expanding and deriving embryonic stem cells in such well-defined, xeno-free culture media. In addition, the present invention provides methods of differentiating ESCs or EBs formed therefrom for the generation of lineage specific cells.
(FR) L'invention concerne des milieux de culture exempts de xéno, bien définis, comprenant un isoforme TGF-bêta ou la chimère formée entre IL6 et le récepteur IL6 soluble (IL6RIL6) et capables de conserver des cellules souches et, notamment, des cellules souches embryonnaires humaines, dans un état non différentié. L'invention concerne, en outre, des cultures de cellules comprenant les milieux de culture et les cellules souches et des procédés d'extension et de dérivation des cellules souches embryonnaires dans de tels milieux de culture exempts de xéno et bien définis. De plus, l'invention concerne des procédés permettant de différentier des ESC ou EB formés à partir de ceux-ci, aux fins de production de cellules spécifiques de lignées.
