(EN)The present invention discloses a two-steps enzyme method for preparing 7-aminocephalosporanic acid from cephalosporin C, Wherein D-amino acid oxidase used is D-amino acid oxidase mutant of purified yeast Trigonopsis Variabilis, which having a specific activity more than 105% higher than that of parent D-amino acid oxidase. The method has no need of addition of hydrogen peroxide, &bgr; -lactamase inhibitor, catalase inhibitor, catalase and the like commonly used in the prior art. The productivity of the method can reach more than 93%.
(FR)La présente invention concerne un procédé enzymatique en deux étapes pour la préparation d'un acide 7-aminocéphalosporanique à partir de céphalosporine C, selon lequel l’oxydase d’acide D-aminé utilisée est un mutant d’oxydase d’acide D-aminé de levure purifiée Trigonopsis Variabilis qui a une activité spécifique plus de 105 % plus élevée que celle de l’oxydase d’acide D-aminé parente. Le procédé n’a pas besoin de l’addition de peroxyde d’oxygène, d’inhibiteur de &bgr; -lactamase, d’inhibiteur de catalase, de catalase et analogue communément utilisés selon la technique antérieure. (ZH)本发明提供了一种从头孢菌素C制备7-氨基头孢霉烷酸的两步酶法,其中所用的D-氨基酸氧化酶为一种经纯化的三角酵母D-氨基酸氧化酶突变体,其所具有的比活比亲本D-氨基酸氧化酶高105%。本发明的方法不需添加过氧化氢、β-内酰胺酶抑制剂、过氧化氢酶抑制剂、过氧化氢酶等现有方法中常用的试剂,而本发明的方法的产率可以达到93%以上,因而具有步骤简单、成本低、产率高的特点。
