Abstract:
(EN) Hydrogel particles comprise protease recognition means, which is adapted to be digested by a protease and cause a variation in the net charge of the particle. This variation in net charge produces a change in the molecular accessibility of the particle. Embodiments of the invention include positively or negatively charged particles. These particles may comprise charged amino acids bonded to the hydrogel polymer via the recognition means. The charged amino acids repel each other and cause swelling of the particle. Digestion of the recognition means causes loss of the charged amino acid and 'collapse' of the particle. The 'swollen' form of the particle may take-up a large molecular which is then entrapped on 'collapse' of the particle. In further embodiments of the invention, the particle comprises oppositely charged amino acid residues flanking the protease recognition means to give an overall neutral particle. Digestion of the protease recognition means causes loss of one of the charged species to leave an overall charged particle which swells and is capable of releasing its contents.
(FR) L'invention concerne des particules d'hydrogel qui comprennent un moyen de reconnaissance de protéase, destiné à être digéré par une protéase et à provoquer une variation au niveau de la charge nette de la particule. Cette variation au niveau de la charge nette produit un changement dans l'accessibilité moléculaire de la particule. Des modes de réalisation selon l'invention comprennent des particules chargées positivement ou négativement. Ces particules peuvent comprendre des acides aminés chargés liés au polymère hydrogel par le biais du moyen de reconnaissance. Les acides aminés chargés se repoussent les uns les autres et entraînent le gonflement de la particule. La digestion du moyen de reconnaissance provoque la perte de l'acide aminé chargé et l''affaissement' de la particule. La forme 'gonflée' de la particule peut prendre un poids moléculaire important qui est alors piégé lors de l''affaissement' de la particule. Dans d'autres modes de réalisation, la particule comprend des résidus d'acides aminés chargés de manière opposée flanquant le moyen de reconnaissance de la protéase pour donner une particule globale neutre. La digestion du moyen de reconnaissance de la protéase entraîne la perte d'une des espèces chargées pour laisser une particule chargée globale qui gonfle et qui est capable de libérer son contenu.
