Abstract:
(EN) Lentivector constructs for expression of recombinant proteins, polypeptides or fragments thereof and methods of making the same are described. The lentivectors typically have a self-processing cleavage sequence between a first and second protein or polypeptide coding sequence allowing for expression of a functional protein or polypeptide under operative control of a single promoter and may further include an additional proteolytic cleavage sequence which provides a means to remove the self-processing cleavage sequence from the expressed protein or polypeptide. The vector constructs find utility in methods relating to enhanced production of biologically active proteins, such as immunoglobulins or fragments thereof in vitro and in vivo.
(FR) L'invention porte sur des lentivecteurs chimères exprimant des protéines et des polypeptides de recombinaison, ou leurs fragments, et sur leurs méthodes d'obtention. Les lentivecteurs comportent normalement une séquence de clivage d'auto-traitement comprise entre une première et une deuxième séquence de codage de protéine ou de polypeptide permettant l'expression d'une protéine fonctionnelle ou d'un polypeptide fonctionnel sous le contrôle opérationnel d'un unique promoteur. Les lentivecteurs peuvent en outre comporter une séquence de clivage protéolytique additionnelle constituant un moyen d'éliminer la séquence de clivage d'auto-traitement des protéines ou polypeptides exprimés. Lesdits vecteurs chimères s'avèrent utiles pour accroître la production de protéines biologiquement actives telles que des immunoglobulines ou leurs fragments t in vitro et in vivo.
