Abstract:
(EN) Disclosed is a method for isolating a pluripotent stem cell which can be differentiated into at least a myocardial cell to regenerate the cardiac muscle, at a high purity. A pluripotent stem cell that can be differentiated into at least a myocardial cell to regenerate the cardiac muscle can be isolated at a high purity by performing the steps of: (i) collecting a skeletal muscle tissue from a mammal and enzymatically treating the skeletal muscle tissue to prepare a cell derived from the skeletal muscle tissue; (ii) cultivating the cell in a culture medium containing an epidermal growth factor and a fibroblast growth factor; and (iii) selecting and separating a colony that is floating in the culture medium.
(FR) La présente invention concerne un procédé permettant d’isoler une cellule souche pluripotente qui présente les propriétés de pouvoir se différencier en au moins une cellule myocardique pour régénérer le muscle cardiaque, à un degré élevé de pureté. Une cellule souche pluripotente qui présente les propriétés de pouvoir se différencier en au moins une cellule myocardique pour régénérer le muscle cardiaque peut être isolée à un degré élevé de pureté en effectuant les étapes de : (i) prélever un tissu du muscle squelettique sur un mammifère et en traitant, à l’aide d’enzymes, le tissu du muscle squelettique pour préparer une cellule dérivée dudit tissu ; (ii) cultiver la cellule dans un milieu de culture qui contient un facteur de croissance épidermique et un facteur de croissance des fibroblastes ; et (iii) sélectionner et séparer une colonie flottant dans le milieu de culture.
(JA)  本発明の目的は、少なくとも心筋細胞に分化して心筋を再生できる多能性幹細胞を高純度で単離する技術を提供することである。  (i)ほ乳動物から骨格筋組織を採取し、得られた骨格筋組織を酵素処理することにより骨格筋組織由来細胞を得る工程、(ii)得られた骨格筋組織由来細胞を、上皮細胞増殖因子及び繊維芽細胞成長因子を含む培地において培養する工程、及び(iii)培養液中に浮遊しているコロニーを選択し、分離する工程を行うことによって、少なくとも心筋細胞に分化して心筋を再生できる多能性幹細胞を高純度で単離することができる。
