Abstract:
(EN) A method of co-formulating two or more pharmaceutically active compounds into a particulate product including contacting a dense, supercritical or near-critical fluid With a suspension of a first active compound in a medium that is miscible with the dense fluid and a solution of a second active compound in a solvent that is miscible with the dense fluid. The product may be the first active agent coated with the second active agent. The method may use coaxial nozzles for the fluid, media or solvents. The method may be applied to production of dry powders for inhalation including beta-agonists and corticosteroids. Apparatus for use in the method comprising means for streams of solvents, media and dense fluids to enter a precipitation chamber at substantially the same point and means for collection of particles under gravity in one or more collection chambers.
(FR) La présente invention concerne un procédé consistant à co-formuler au moins deux composés pharmaceutiquement actifs pour former un produit particulaire, qui comprend la mise en contact d’un fluide supercritique ou proche de critique dense ave une suspension d’un premier composé actif dans un milieu miscible avec le fluide dense et une solution d’un second composé actif dans un solvant miscible avec le fluide dense. Le produit peut être le premier agent actif enrobé avec le second agent actif. Ce procédé peut utiliser des buses coaxiales pour le fluide, les milieux ou les solvants. Ce procédé peut être appliqué à la production de poudres sèches pour inhalation, comprenant notamment des bêta-agonistes et des corticostéroïdes. L’appareil destiné à être utilisé dans le cadre de ce procédé comprend un moyen permettant aux flux de solvants, milieux et fluides denses d’entrer dans une chambre de précipitation sensiblement au même point ainsi qu’un moyen de collecte des particules sous l’effet de la gravité dans une ou plusieurs chambres de collecte.
