Abstract:
(EN) The present invention provides novel technologies for producing and screening fusion proteins on the surface of filamentous phage. In particular, a single vector can be used for generating cell and phage libraries containing a given series of protein sequences fused to either one or other of two phage coat proteins. This approach simplifies and improves the efficiency of the subsequent phage display-based selection of protein-binding molecules having therapeutic or diagnostic utility, such as antibodies, peptides, or epitope-binding regions.
(FR) La présente invention concerne des nouvelles technologies de production et de criblage de protéines de fusion sur la surface d'un phage filamenteux. Plus particulièrement, il est possible d'utiliser un seul vecteur pour produire des bibliothèques de cellules et de phages contenant une série donnée de séquences de protéines fusionnées à l'une ou l'autre des deux protéines protéines de coque phage. Cette méthode simplifie et améliore l'efficacité d'une sélection ultérieure, par présentation de molécules par les phages, de molécules de liaison de protéines présentant une utilité thérapeutique ou diagnostique telles qu'anticorps, peptides ou régions de liaison d'épitope.
