Abstract:
(EN) Provided is a novel approach for generating oligonucleotide probes and the use of these probes in gene expression profiling, by hybridization to test oligonucleotides on arrays or beads. This approach involves labeling of the complement oligonucleotide probes using a mixture of dye or hapten labeled-ddNTPs in solution. The labeled oligonucleotide probes are then used to hybridize to the test oligonucleotides on the solid support. Success in hybridization is monitored by associated signal on the solid support. This approach greatly reduces hybridization time, due to the simplification of the probe content. It is especially useful when analyzing a small number of genes, such as a signature set of genes for a disease or condition.
(FR) L'invention concerne un nouveau procédé de production de sondes oligonucléotidiques et l'utilisation desdites sondes dans le profilage de l'expression génique, par leur hybridation à des oligonucléotides à l'essai sur des puces ou des billes. Selon le procédé de l'invention, on marque des sondes oligonucléotidiques complémentaires au moyen d'un mélange de colorant ou de ddNTP hapténisés en solution. On procède alors à l'hybridation des sondes oligonucléotidiques marquées avec les oligonucléotides à l'essai sur le support solide. On contrôle le succès de l'hybridation au moyen d'un signal associé sur le support solide. Le procédé de l'invention réduit significativement le temps d'hybridation en raison de la simplification du contenu de la sonde. Le procédé est particulièrement utile lorsqu'on analyse un petit nombre de gènes, par exemple un ensemble de signatures géniques correspondant à une maladie ou à un état pathologique.
