Abstract:
(EN) The present invention provides recombination based methods for assembling nucleic acids. In certain aspects the present invention provides hierarchical assembly methods for producing genome sized polynucleotide constructs. The methods may be used for assembling large polynucleotide constructs, for synthesizing synthetic genomes, or for introducing a plurality of nucleotide changes throughout the genome of an organism. In another aspect, the invention provides cells having increased genomic stability. For example, cells comprising alterations in at least a substantial portion of the transposons in the genome are provided.
(FR) La présente invention a trait à des procédés basés sur la recombinaison pour l'assemblage d'acides nucléiques. Dans certains aspects, la présente invention a trait à des procédés d'assemblage hiérarchique pour la production de constructions polynucléotidiques de taille génomique. Les procédés peuvent être utilisés pour l'assemblage de grandes constructions polynucléotidiques, pour la synthèse de génomes synthétiques, ou pour l'introduction d'une pluralité de modifications dans le génome entier d'un organisme. Dans un autre aspect, l'invention a trait à des cellules présentant une stabilité génomique accrue, par exemple, des cellules comportant des modifications dans au moins une portion substantielle de transposons dans le génome.
