Abstract:
(EN) The object is to construct a DNA replication reaction system which is excellent in general versatility and is easy to use. Disclosed is a monomer of PCNA which is a factor involved in DNA replication. The monomer has such an amino acid sequence that, when the monomer is linked to other monomer to form a polymer in which the N-terminus of the monomer and the C-terminus of the other monomer forms the boundary between these monomers, the amino acid residues of these monomers at the site at which an intermolecular interaction between these monomers is formed at the interface are those amino acids which repel each other by virtue of their electrical charges.
(FR) L’objet de la présente invention consiste à construire un système de réaction de réplication d’un ADN, présentant une excellente versatilité générale et facile d’emploi. L’invention se rapporte à un monomère d’antigène nucléaire de prolifération cellulaire (PCNA), qui est un facteur impliqué dans la réplication de l’ADN. Le monomère a une séquence d’acide aminé telle que lorsque le monomère est relié à un autre monomère pour former un polymère dans lequel l’extrémité N du monomère et l’extrémité C de l’autre monomère forment une frontière entre ces monomères, les résidus d’acide aminé de ces monomères au niveau du site sur lequel une interaction intermoléculaire entre ces monomères est formée à l’interface, sont les acides aminés qui se repoussent les uns les autres en raison de leurs charges électriques.
(JA)  汎用性に優れ、使いやすいDNA複製反応系を構築することを課題とする。DNA複製に関与する因子の一つであるPCNAの単量体のアミノ酸配列を、単量体のN末端側領域と他の単量体のC末端側領域とが界面となって多量体を形成した場合に、各単量体の界面領域で単量体相互の分子間相互作用を形成する部位のアミノ酸残基が、電荷的に反発しあうアミノ酸残基で構成されるように調製する。
