Abstract:
(EN) A method of detecting a molecule associated with viability of one or more cells or organisms in a sample comprises the initial step of contacting the sample with an enzyme, which enzyme is capable of adding or removing a chemical moiety to or from a nucleic acid molecule in the presence of the molecule associated with viability of the one or more cells or organisms. This thereby generates a novel detectable nucleic acid molecule. The next step involves detecting the presence of the molecule associated with viability of the one or more cells or organisms by detecting the novel nucleic acid molecule generated only in the presence of the molecule associated with viability of the one or more cells or organisms. A most preferred molecule associated with viability is ATP, although NAD may also be detected. A preferred enzyme for use in the methods is ligase. The method has numerous applications, in particular in monitoring viability of cells, toxicology testing and determining whether there is contamination in a sample or on a surface. Kits are also provided for carrying out the methods.
(FR) L'invention concerne un procédé de détection, dans un échantillon, d'une molécule associée à la viabilité d'une ou de plusieurs cellules ou organismes. Le procédé comporte les étapes consistant à: mettre l'échantillon en contact avec une enzyme, cette enzyme permettant d'ajouter ou d'enlever une fraction chimique à une molécule ou de celle-ci, en présence de la molécule associée à la viabilité d'une ou de plusieurs cellules ou organismes, cette étape initiale produisant une nouvelle molécule d'acide nucléique détectable; détecter la présence de la molécule associée à la viabilité d'une ou de plusieurs cellules ou organismes en détectant la nouvelle molécule d'acide nucléique produite seulement en présence de la molécule associée à la viabilité d'une ou de plusieurs cellules ou organismes. Idéalement, la molécule associée à la viabilité est l'ATP, mais on peut aussi détecter l'ADN. L'enzyme préférée utilisée dans les procédés est la ligase. Ce procédé comporte de nombreuses applications, en particulier dans la surveillance de la viabilité de cellules, les essais toxicologiques et pour déterminer la présence d'une contamination dans un échantillon ou sur une surface. Des trousses pour mettre en oeuvre les procédés sont également prévues.
