Abstract:
(EN) An ex vivo method of expanding animal cells whose differentiation state is controllable by modulating TGF-&bgr; signaling includes the steps of: (a) providing an animal subject having cells with a first phenotype; (b) isolating the cells from the animal subject; (c) placing the cells in an ex vivo culture system including a culture vessel having at least one surface and a medium in contact with the at least one surface, the medium being essentially free of intact amniotic membrane and feeder cells; and (d) culturing the cells in the medium under conditions which downregulate TGF-&bgr; signaling in the cells and allow the cells to proliferate while maintaining the first phenotype.
(FR) L'invention concerne une méthode d'expansion ex vivo de cellules animales dont l'état de différenciation est régulable par modulation de la signalisation du TGF-$g(b). Cette méthode consiste (a) à faire appel à un sujet animal possédant des cellules présentant un premier phénotype, (b) à isoler les cellules de ce sujet animal, (c) à placer les cellules dans un système de culture ex vivo comprenant un récipient de culture possédant au moins une surface et un milieu en contact avec cette surface, le milieu étant essentiellement exempt de membrane amniotique intacte et de cellules nourricières, et (d) à cultiver les cellules dans ce milieu dans des conditions provoquant une régulation à la baisse de la signalisation du TGF-$g(b) dans les cellules et permettant aux cellules de proliférer tout en conservant le premier phénotype.
