(EN)A test is performed on the effects of an interferon on the expression of HLA-A and the cytotoxicity caused by an anti-HLA class I antibody in a cell, using a 2D7-DB antibody as the anti-HLA class I antibody and an IM-9 cell strain as the cell to be tested. As a result, it is demonstrated that the expression of HLA-A can be up-regulated by IFN&agr; and IFNϝ. Further, a WST-8 assay performed using an interferon and an anti-HLA class I antibody in combination to examine the cell viability demonstrates that cell death activity can be increased by the combination of 2D7-DB and IFN&agr; or IFNϝ.
(FR)La présente invention concerne un test évaluant les effets d’un interféron sur l’expression de l’HLA-A et la cytotoxicité induite par un anticorps anti-HLA de classe I dans une cellule, utilisant un anticorps 2D7-DB comme anticorps anti-HLA de classe I et une souche cellulaire IM-9 comme cellule à tester. Ce test démontre que l’expression de l’HLA-A peut être régulée de manière positive par IFN&agr; et IFNϝ. En outre, un test WST-8 réalisé en utilisant une association à base d’un interféron et d’un anticorps anti-HLA de classe I pour déterminer la viabilité cellulaire démontre que l’activité de mort cellulaire peut être accrue par l’association comprenant 2D7-DB et IFN&agr; ou IFNϝ.
(JA)インターフェロンが細胞のHLA-Aの発現および抗HLAクラスI抗体による細胞障害にどのような効果を及ぼすかについて、抗HLAクラスI抗体として2D7-DB抗体を用い、IM-9細胞株を対象に試験を行った結果、IFNαまたはIFNγにより、HLA-Aの発現が上方制御されることが実証された。さらに、WST-8アッセイにより、インターフェロンと抗HLAクラスI抗体を併用した場合の細胞生存度を検討ところ、2D7-DBとIFNαまたはIFNγとの組み合わせにより、細胞死活性が増大することがわかった。
