Abstract:
(EN) Compositions are described for detecting binding of an analyte to a binding partner attached to a nucleic acid hybridization switch probe that includes first and second arm sequences and a support sequence that is at least partially complementary to both arm sequences, allowing the probe under hybridization conditions to form a first conformation in the absence of the analyte and to form a second conformation in the presence of the analyte, and a label associated with the probe that produces a signal that indicates the conformation of the probe. Methods are described for detecting an analyte that forms a specific binding pair with the binding partner attached to the hybridization switch probe, thereby changing the probe from a first to a second conformation that results in a detectable signal that indicates the presence of the analyte in the sample.
(FR) Les compositions décrites dans la présente invention sont destinées à détecter la liaison d’une substance à analyser à un partenaire de liaison fixé à une sonde à commutation d’hybridation d’acide nucléique comprenant les première et deuxième séquences de bras et une séquence de support au moins partiellement complémentaire aux deux séquences de bras, permettant à la sonde, dans des conditions d’hybridation, de former une première conformation en l’absence de la substance à analyser et de former une deuxième conformation en présence de la substance à analyser, et un traceur accompagnant la sonde qui émet un signal pour indiquer la conformation de la sonde. Les procédés décrits dans la présente invention sont destinés à détecter une substance à analyser qui forme une paire de liaison spécifique avec le partenaire de liaison fixé à la sonde à commutation d’hybridation, faisant ainsi passer la sonde d’une première conformation à une deuxième conformation, ce qui déclenche un signal détectable indiquant la présence de la substance à analyser dans l’échantillon.
