(EN)The invention describes a method for identifying modified versions of a foreign recombinant peptide, polypeptide or protein, expressed from a cloned gene, which are produced and maintained in a host cell at high levels and which are of reduced toxicity, such that genetic stability of the recombinant DNA is increased. This is achieved by a dual selection process whereby the modified protein is tested both (i) for its ability to accumulate and (ii) for its ability to cause reduced host stress.
(FR)L'invention porte sur une méthode d'identification de versions modifiées d'un peptide, d'un polypeptide ou d'une protéine de recombinaison exprimés par un gène cloné, et qui sont produits et conservés dans une cellule hôte à haut niveau et qui présentent une toxicité réduite d'où un accroissement de la stabilité génétique de l'ADN de recombinaison. Cela s'obtient par un double processus de sélection selon lequel on teste (i) la capacité d'accumulation et (ii) la capacité à produire un stress réduit dans la cellule hôte, de la protéine modifiée.
