(EN)The present invention provides compositions and methods for the production of differentiated mammalian cells. More particularly, the present invention provides cellular differentiation methods employing culturing the cells on a feeder layer or under feeder-free conditions in cell culture and further contacting the cells with an inhibitor of the PI3-kinase pathway and a member of the TGFb family for the generation of differentiated mammalian cells from pluripotent mammalian stem cells. Preferably, the differentiated cell is selected from the group consisting of a mesendodermal cell, a mesodermal cell, and an endodermal cell.
(FR)L'invention concerne des compositions et des procédés pour produire des cellules différenciées de mammifère. L'invention concerne en particulier des procédés de différenciation cellulaire selon lesquels des cellules sont cultivées sur une couche nourricière ou dans des conditions exemptes d'alimentation en une culture cellulaire, les cellules étant mises en contact avec un inhibiteur de la voie de la PI3-kinase et avec un membre de la famille TGF$g(b) pour générer des cellules différenciées de mammifère à partir de cellules souches pluripotentes de mammifère. La cellule différenciée est de préférence sélectionnée dans le groupe comprenant une cellule méso-endodermique, une cellule mésodermique et une cellule endodermique.
