Abstract:
(EN) It is intended to produce a target protein at a high efficiency with the use of the action of FKBP type PPIase which shows a sufficient activity in a region from low temperature to ordinary temperature in usual biochemical environment. Owing to the action of FKBP type PPIase originating in a non-thermophilic archaebacterium belonging to methane-producing bacteria, a target protein is obtained as a normal protein. By a fused protein composed of FKBP type PPIase with the target protein or the coexpression of the FKBP type PPIase with the target protein, the target protein can be properly folded. By using a vector which can express the fused protein, the target protein can be produced at a high efficiency.
(FR) L'invention concerne la production d'une protéine cible à haut rendement par l'action de la PPIase de type FKBP qui présente une activité suffisante dans une région de basse température à température ordinaire dans un milieu biochimique habituel. Compte tenu de l'action de la PPIase de type FKBP prenant naissance dans une archaébactérie non thermophile appartenant aux bactéries produisant du méthane, on obtient une protéine cible comme protéine normale. Par une protéine fusionnée, composée de la PPIase de type FKBP ou la coexpression de cette dernière avec la protéine cible, on peut plier la protéine cible de manière appropriée. En utilisant un vecteur pouvant exprimer la protéine cible, on peut produire la protéine cible à haut rendement.
(JA)  低温から常温域でかつ通常の生化学的な環境下でも十分な活性を示すFKBP型PPIaseの作用により、目的タンパク質を高効率で製造することを課題とする。  メタン生成菌に属する常温性古細菌由来のFKBP型PPIaseの作用により、目的タンパク質を正常型タンパク質として取得する。該FKBP型PPIaseと目的タンパク質との融合タンパク質、又は該FKBP型PPIaseと目的タンパク質の共発現により目的タンパク質は正しく折り畳まれる。該融合タンパク質を発現することができるベクターによって、目的タンパク質は高効率で製造される。
