Abstract:
(EN) Provided is a method for characterising an analyte by matrix assisted laser desorption ionisation (MALDI) mass spectrometry, which method comprises: (a) labelling the analyte with a light-absorbing label that absorbs light at a pre-determined frequency, to form a labelled analyte; (b) embedding the labelled analyte in a matrix formed from at least one compound that absorbs light, to form an embedded labelled analyte; (c) desorbing the embedded labelled analyte by exposing it to light having the pre-determined frequency, to form a desorbed analyte; and (d) detecting the desorbed analyte by mass spectrometry to characterise the analyte; wherein the light absorbing label comprises a fluorophore moiety, and wherein prior to detecting by mass spectrometry, the analyte is selected for detection on the basis of its fluorophore moiety.
(FR) L'invention concerne un procédé permettant de caractériser une substance à analyser par spectrométrie de masse à désorption-ionisation par impact laser assistée par matrice (MALDI). Ce procédé consiste : (a) à marquer la substance à analyser à l'aide d'un marqueur d'absorption de lumière qui absorbe la lumière à une fréquence prédéterminée, pour former une substance à analyser marquée ; (b) à intégrer la substance à analyser marquée dans une matrice formée à partir d'au moins un composé qui absorbe la lumière, pour former une substance à analyser marquée intégrée ; (c) à désorber ladite substance à analyser marquée intégrée par exposition de cette dernière à la lumière présentant une fréquence prédéterminée pour former une substance à analyser désorbée ; et (d) à détecter la substance à analyser désorbée par spectrométrie de masse de manière à caractériser la substance à analyser. Dans ledit procédé, le marqueur d'absorption de lumière comprend un fragment flurophore ; et préalablement à la détection par spectrométrie de masse, la substance à analyser est sélectionnée pour être détectée sur la base de son fragment fluorophore.
