(EN)The invention relates to methods and compositions for causing the selective targeting and killing of tumor cells. Through ex vivo gene therapy protocols tumor cells are engineered to express AN &agr; (1,3) galactosyl epitope. The cells are then irradiated or otherwise killed and administered to a patient. The &agr; galactosyl epitope causes opsonization of the tumor cell enhancing uptake of the opsonized tumor cell by antigen presenting cells which results in enhanced tumor specific antigen presentation. The animal’s immune system thus is stimulated to produce tumor specific cytotoxic cells and antibodies which will attack and kill tumor cells present in the animal.
(FR)L'invention concerne des méthodes et des compositions permettant de cibler et de détruire sélectivement des cellules tumorales. A l'aide de protocoles de thérapie génique ex vivo, on conçoit par génie génétique des cellules tumorales exprimant un épitope de $g(a) (1,3) galactosyle. Ces cellules sont ensuite irradiées ou tuées et administrées à un patient. Un épitope de galactosyle provoque une opsonisation de la cellule tumorale, ce qui favorise le captage de la tumeur opsonisée par les cellules porteuses de l'antigène et favorise la présentation d'antigènes spécifiques de la tumeur. Le système immunitaire ainsi stimulé de l'animal produit des cellules et des anticorps cytotoxiques de la tumeur qui vont attaquer et détruire les cellules cancéreuses présentes chez l'animal.
