A method for isolating nucleic acid molecules having a repeating nucleotide sequence or a homopolymeric nucleotide sequence, e.g. a poly A stretch, is described. In particular, the method uses oligomeric capture probes spiked with various amounts of locked nucleic acid (LNA). The invention further describes methods for the isolation of RNA molecules, for example polyadenylated mRNA molecules, which overcome the problems of rapid RNA degradation during isolation and analysis of such nucleic acid molecules. This is of major clinical and diagnostic importance, especially when dealing with RNA viruses, such as retroviruses or when analyzing rare or low-abundant mRNAs or mRNAs from biopsies or tissues enriched with RNases.

L'invention concerne un procédé d'isolation de molécules d'acide nucléique présentant une séquence nucléotidique répétitive ou une séquence nucléotidique homopolymère, par exemple une extension poly A. Plus précisément, le procédé utilise des sondes de capture oligomères dont les pointes comprennent diverses quantités d'acide nucléique verrouillé (LNA). L'invention concerne également des procédés d'isolation de molécules d'ARN, par exemple des molécules d'ARNm polyadénylées, résolvant les problèmes de la dégradation rapide de l'ARN pendant l'isolation et l'analyse de telles molécules d'acide nucléique. Cela a une importance clinique et diagnostique capitale, surtout lors de la manipulation de virus ARN, tels que des rétrovirus ou lors de l'analyse d'ARNm rares ou à faible abondance ou d'ARNm provenant de biopsies ou de tissus enrichis en RNases.
