Abstract

A method for preparing a nucleic acid component of a sample for amplification includes contacting the sample with a porous support that deactivates a nucleic acid amplification inhibitor component of the sample and directing a fluid through the porous support, whereby the nucleic acid component of the sample is directed through at least a portion of the porous support and is separated from the support, thereby preparing the nucleic acid component for amplification. The sample is separated from raw sample components through means that include a magnetic substrate. An apparatus suitable for conducting the method of the invention also is described.

L'invention concerne un procédé de préparation d'une composante acide nucléique d'un échantillon aux fins d'amplification. Ce procédé consiste à mettre en contact l'échantillon avec un support poreux qui désactive une composante de l'échantillon, composante inhibitrice de l'amplification de l'acide nucléique, et à diriger un fluide à travers le support poreux, la composante acide nucléique de l'échantillon étant dirigée à travers au moins une partie du support poreux et étant séparée du support, ce qui permet d'obtenir la composante acide nucléique destinée à l'amplification. L'échantillon est séparé des composantes d'échantillon brutes par un moyen qui comprend un substrat magnétique. L'invention concerne également un appareil convenant à la mise en oeuvre du procédé selon l'invention.
