The present invention provides methods and kits for identifying an increased risk of developing cancer in a subject. The methods include analyzing a first biological sample, such as a blood sample, from the subject for loss of imprinting of the IGF2 gene. According to the methods a loss of imprinting is indicative of an increased risk of developing cancer. The method can include analyzing genomic DNA from the sample for altered methylation of the IGF2 gene. The altered methylation for example includes hypomethylation of a differentially methylated region of IGF2, corresponding to SEQ ID NO: 1 or a polymorphism thereof. The method can be performed on a subject having no apparent or suspected hyperproliferative disorder such as cancer.

Cette invention porte sur des méthodes et des trousses permettant d'identifier un risque accru de développement d'un cancer chez un sujet. Ces méthodes consistent à analyser un premier échantillon biologique tel qu'un échantillon sanguin prélevé sur le sujet afin d'y déceler une perte de marquage du gène IGF2. Selon les méthodes de cette invention, une perte de marquage indique un risque accru de développement d'un cancer. Cette méthode peut consister à analyser un ADN génomique de l'échantillon pour déceler une modification de la méthylation du gène IGF2. Cette modification de méthylation comprend par exemple l'hypométhylation d'une région à méthylation différentielle de l'IGF2, correspondant à la séquence SEQ ID NO:1 ou à un polymorphisme de celle-ci. Cette méthode peut être mise en oeuvre chez un sujet ne présentant aucun trouble hyperprolifératif apparent ou présumé tel qu'un cancer.
