Abstract
(EN) A process is described for the preparation of (RR,SS˜2-dimethylaminomethyl-1-(3-methoxyphenyl) cyclohexanol hydrochloride (hereinafter 'tramadol hydrochloride'), which process comprises reacting a mixture of (RR,SS)- and (RS,SR)-2-dimethylaminomethyl-l-(3-methoxyphenyl) cyclohexanol with HCI in the presence of a catalytic amount of water. The process can be carried out in the absence of 1,4-dioxane or other category 1 carcinogenic solvent, and in one step, providing commercial advantages of simplicity, safety and purity. Tramadolhydrochloride is a known non-narcotic analgesic.
(FR) La présente invention a trait à un procédé de préparation de chlorhydrate de (RR,SS)- 2-diméthylaminométhyl-1-(3-méthoxyphényl) cyclohexanol (ci-après désigné chlorhydrate de tramadol), lequel procédé comporte la réaction d'un mélange de (RR,SS)- et (RS,SR)-2- diméthylaminométhyl-1-(3-méthoxyphényl) cyclohexanol avec du chlorhydrate en présence d'une quantité catalytique d'eau. Le procédé peut être effectué en l'absence de 1,4-dioxane ou autre solvant carcinogène de catégorie 1, et en une seule étape, procurant ainsi les avantages commerciaux de la simplicité, de la sécurité et de la pureté. Le chlorhydrate de tramadol est un analgésique non narcotique connu.
