(EN)It is intended to provide a method of efficiently constructing a germline chimeric animal and a method of conveniently constructing a heterozygous animal and a homozygous animal. More specifically speaking, embryonic stem cells (ES cells) are injected into an early embryo (i.e., a blastocyst) which cannot form germ cells due to a genetic cause to thereby construct a chimeric animal carrying germ cells originating in the ES cells thus injected. By crossing male and female heterozygotes obtained from the chimeric animal, a homozygote can be obtained. The chimeric animal thus obtained is highly useful in analyzing gene functions, etc.
(FR)L'invention concerne un procédé de construction efficace d'animal chimère à lignée germinale et un procédé de construction pratique d'animal hétérozygote et d'animal homozygote. De façon plus spécifique, des cellules souches embryonnaires sont injectées dans un jeune embryon (c'est à dire un blastocyste) qui ne peut génétiquement former de cellules germinales, afin de construire un animal chimère comportant des cellules germinales provenant des cellules souches embryonnaires injectées. Par croisement de mâle et de femelle hétérozygotes obtenus à partir de l'animal chimère, il est possible d'obtenir un homozygote. L'animal chimère ainsi obtenu est très utile, en particulier dans l'analyse des fonctions génétiques.
