Die vorliegende Erfindung betrifft ein Verfahren zum Nachweis Epitop−spezifischer T−Zellen und von Zielepitopen reaktiver T−Zellen. Die vorliegende Erfindung betrifft ferner Vektoren, umfassend einen spezifisch in antigenpräsentierenden Zellen durch den epitop−spezifischen Kontakt mit einer T−Zelle induzierbaren ersten Promotor, eine mit diesem ersten Promotor funktionell verbundene Nukleinsäure kodierend ein Markergen, einen in antigenpräsentierenden Zellen konstitutiven zweiten Promotor, und eine mit diesem zweiten Promotor funktionell verbundene Nukleinsäure. Die vorliegende Erfindung betrifft ferner antigenpräsentierende Zellen, transduziert mit den erfindungsgemäßen Vektoren.

The invention relates to a method for assaying epitope-specific T cells and target epitopes of reactive T cells. The present invention further relates to vectors, comprising a first promoter that can be specifically induced in antigen-presenting cells by the epitope-specific contact with a T cell, a nucleic acid functionally linked with said first promoter and encoding a marker gene, a second promoter that is constitutive in antigen-presenting cells, and a nucleic acid that is functionally linked with said second promoter. The present invention further relates to antigen-presenting cells that are transduced with the vectors according to the invention.

La présente invention concerne un procédé de détection de lymphocytes T spécifiques à l'épitope et d'épitopes cibles de lymphocytes T réactifs. La présente invention concerne également des vecteurs contenant un premier promoteur pouvant être induit spécifiquement dans les cellules présentant un antigène par contact spécifique à l'épitope avec un lymphocyte T, un acide nucléique fonctionnellement lié à ce premier promoteur et codant un gène marqueur, un deuxième promoteur constitutif dans les cellules présentant un antigène, et un acide nucléique fonctionnellement lié à ce deuxième promoteur. La présente invention concerne également des cellules présentant un antigène et transduites par les vecteurs selon l'invention.
