Abstract

Crystallization apparatus (20) and methods which allow for fast screening and determination of protein crystallization conditions with small protein quantities and rapid crystallization. The apparatus comprise a first region comprising a first nucleation catalyst material (36) and a second region comprising a second nucleation catalyst material (38), with the first and second regions positioned adjacent to each other and configured to support at least one crystal, and with the first region having a variation in a nucleation property of the first nucleation catalyst material (36), in the first region. The crystal may be supported at an interface of the adjacent regions. The methods comprise providing a first region of a first nucleation catalyst material (36) and a second region of a second nucleation catalyst material (38) adjacent said first region, with the first region having a variation in a nucleation property of the first nucleation catalyst material (36), exposing the first and second regions to a solution of a selected molecule, and growing at least one crystal of the molecule in association with the first and second regions.

L'invention concerne un appareil et des procédés de cristallisation permettant de cribler et de déterminer rapidement des conditions de cristallisation de protéines au moyen de petites quantités de protéines et d'une cristallisation rapide. Ledit appareil comprend une première zone comportant une première matière catalyseur de nucléation et une seconde zone comportant une seconde matière catalyseur de nucléation, la première et la seconde zone étant agencées de manière mutuellement adjacente et configurées afin de soutenir au moins un cristal, et la première zone présentant une variation dans une propriété de nucléation de la première matière catalyseur de nucléation de la première zone. Ledit cristal peut être supporté au niveau d'une interface des zones adjacentes. Les procédés consistent à se munir d'une première zone d'une première matière catalyseur de nucléation et d'une seconde zone d'une seconde matière catalyseur de nucléation adjacente à ladite première zone, la première zone présentant une variation dans une propriété de nucléation de la première matière catalyseur de nucléation, à exposer la première et la seconde zone à une solution d'une molécule sélectionnée, et à générer au moins un cristal de la molécule associé à la première et à la seconde zone.
