The present invention provides a method of preparing a nucleic acid sample comprising template nucleic acid and synthetic nucleic acid for analysis wherein prior to analysis the nucleic acid sample is treated with a substance which selectively cleaves the template nucleic acid without substantially cleaving the synthetic nucleic acid. The invention further provides a method for improving the analysis of capillary-based DNA sequencing reactions, amplification reactions, and/or transcription reactions, wherein after the reaction, the nucleic acid sample comprising template nucleic acid and synthetic nucleic acid is treated with a substance which selectively cleaves the template without substantially cleaving the synthetic nucleic acid.

L'invention concerne un procédé permettant de préparer un échantillon d'acide nucléique comprenant une matrice d'acide nucléique et un acide nucléique de synthèse pour une analyse. Ce procédé consiste à traiter avant l'analyse l'échantillon d'acide nucléique avec un produit qui provoque un clivage sélectif de la matrice d'acide nucléique sans clivage notable de l'acide nucléique synthétique. L'invention concerne en outre un procédé permettant d'améliorer l'analyse des réactions de séquençage, des réactions d'amplification et/ou des réactions de transcription d'ADN au moyen de capillaires. Ce procédé consiste à traiter après la réaction l'échantillon d'acide nucléique comprenant la matrice d'acide nucléique et l'acide nucléique de synthèse au moyen d'un produit entraînant un clivage sélectif de la matrice sans clivage notable de l'acide nucléique de synthèse.
