(EN)It is intended to provide a method of screening a remedy for heart failure, a remedy for hypertension, etc. characterized by inhibiting the binding of GPK protein having a pH domain or its peptide fragment to G beta gamma protein to thereby directly confirm G protein-coupled receptor desensitization inhibition. According to this screening method, a large amount of a compound can be quickly and conveniently screened and the thus obtained compound, which alters the binding of GPK protein to G bata gamma protein, is useful as a remedy for heart failure, hypertension, etc. By inhibiting the binding of GRK2 to G beta gamma protein, the phosphorylation effect of GRK on $g(b)AR can be regulated and the desensitization can be inhibited, thereby enabling the development of drugs based on a novel function mechanism.
(FR)L'invention porte sur un procédé de criblage d'un remède contre l'insuffisance cardiaque, l'hypertension, etc., inhibant la fixation d'une protéine GPK, présentant un domaine pH ou ses fragments, sur la protéine $g(b)$g(g), ce qui confirme directement l'inhibition de la désensibilisation du récepteur lié à la protéine G. Selon ledit procédé une importante quantité du composé peut être rapidement et correctement criblée, et le composé ainsi obtenu qui modifie la fixation de la protéine GPK sur la protéine $g(b)$g(g) peut être utilisé comme remède contre l'insuffisance cardiaque, l'hypertension, etc. En empêchant la fixation de la GRK2 sur la protéine $g(b)$g(g), on peut réguler l'effet de la phosphorylation de la GRK sur la $g(b)AR et inhiber la désensibilisation.
