Methods for engineering a target activated biosensor are provided. Biosensors comprise a plurality of nucleic acid sensor molecules labeled with a first signaling moiety and a second signaling moiety. The nucleic acid sensor molecules recognizes target molecules which do not naturally bind to DNA. Binding of a target molecule to the sensor molecules triggers a change in the proximity of the signaling moieties which leads to a change in the optical properties of the nucleic acid sensor molecules on the biosensor. Reagents and systems for performing the method are also provided. The method is useful in diagnostic applications and drug optimization.

L'invention concerne des procédés de production d'un biocapteur activé par une cible. Les biocapteurs comportent une pluralité de molécules de détection d'acide nucléique marquées par un premier fragment de signalisation et un second fragment de signalisation. Les molécules de détection d'acide nucléique reconnaissent les molécules cibles qui ne se fixent pas naturellement à l'ADN. La fixation d'une molécule cible aux molécules de détection déclenche un changement à proximité des fragments de signalisation, ce qui provoque un changement dans les propriétés optiques des molécules de détection d'acide nucléique sur le biocapteur. L'invention concerne également des réactifs et des dispositifs de mise en oeuvre dudit procédé. Le présent procédé est utile dans les applications diagnostiques et l'optimisation des médicaments.
