The present inventors have discovered that Mg-chelatase is essential for the growth of Arabidopsis. Specifically, the inhibition of Mg-chelatase CHL,H gene expression in Arabidopsis seedlings results in varying levels of chlorosis (yellowing), significantly reduced growth and developmental abnormalities. Thus, Arabidopsis Mg-chelatase can be used as a target for the identification of herbicides. Accordingly, the present invention provides methods for the identification of compounds that modulate Arabidopsis Mg-chelatase expression or activity, comprising: contacting a compound with a Arabidopsis Mg-chelatase, or a subunit thereof, and detecting the presence and/or absence of binding between said compound and said Mg-chelatase, or detecting a change in Mg-chelatase expression or activity. The methods of the invention are useful for the identification of herbicides and other compounds that can modulate plant growth and development. In addition, the methods of the invention are useful for the identification of compounds that stimulate the expression or function of Mg-chelatase expression or function. Such compounds can be used to promote or manipulate plant growth and development.

L'invention concerne la chélatase de magnésium dont il a été découvert qu'elle est essentielle pour la croissance d'<i>Arabidopsis</i>. De manière plus spécifique, l'inhibition de l'expression du gène CHL H de la chélatase de magnésium dans les plants d'<i>Arabidopsis</i> provoque la modification des niveaux de chlorose (jaunissement), une croissance sensiblement réduite et des anomalies de développement. Ainsi, la chélatase de magnésium d'<i>Arabidopsis</i> peut être utilisée comme cible pour l'identification d'herbicides. La présente invention porte aussi sur des procédés pour identifier des composée qui modulent l'expression ou l'activité de la chélatase de magnésium d'<i>Arabidopsis</i>, comprenant les opérations suivantes : mettre en contact un composé avec la chélatase de magnésium d'<i>Arabidopsis</i> ou avec une sous-unité de celle-ci, et déterminer la présence et/ou l'absence de liant entre ledit composé et ladite chélatase de magnésium, ou bien détecter un changement dans l'expression ou l'activité de la chélatase de magnésium. Ces procédés sont utiles pour identifier des herbicides ou d'autres composés susceptibles de moduler la croissance et le développement d'une plante. Lesdits procédés sont en outre utiles pour l'identification de composés qui stimulent l'expression ou la fonction de la chélatase de magnésium. De tels composés peuvent être utilisés pour favoriser ou manipuler la croissance et le développement d'une plante.
