Abstract

The present invention relates to a zebrafish HuC promoter with its 5'-flanking region which directs the neuron-specific expression of structural genes, a transgenic animal that shows the neuron-specific expression of GFP (green fluorescence protein) under the regulation of the HuC promoter, and a method for screening neurogenesis mutants in zebrafish by use of the transgenic animal. The HuC promoter with its 5'-flanking region can be used in the study of the regulatory mechanism responsible for the differentiation of the nervous system. Additionally, the HuCP-GFP transgenic zebrafish enables the direct identification of neurogenesis and axonogenesis, as well as being a valuable tool for isolating and analyzing neurogenesis mutants in live zebrafish with ease.

L'invention concerne un promoteur HuC de poisson zèbre associé à sa région flanquante 5' qui dirige l'expression spécifique aux neurons de gènes structuraux, un animal transgénique qui présente l'expression spécifique aux neurons de la protéine GFP (protéine verte fluorescente) sous la régulation du promoteur HuC, ainsi qu'un procédé de criblage de mutants de neurogénèse chez les poissons zèbres par utilisation de l'animal transgénique. Le promoteur HuC et sa région flanquante 5' peuvent être utilisés dans l'étude du mécanisme régulatoire responsable de la différenciation du système nerveux. Le poisson zèbre transgénique HuCP-GFP permet, par ailleurs, l'identification directe de la neurogénèse et de l'axonogenèse, et représente un outil précieux pour facilement isoler et analyser les mutants de neurogénèse chez des poissons zèbres vivants.
