Abstract
(EN) The invention relates generally to the area of drug development and more specifically to screening novel compounds for antimicrobial activity. Methods are described for generating mutagenized peptide libraries expressed in colonies of cells. A fluorescent live/dead cell assay combined with a digital imaging spectrophotometer provides a high-throughput solid-phase screening method for colonies or arrayed synthetic libraries. The assay enables screening of antimicrobial peptide activity in at least 105-106 colonies per experiment. These methods generate and identify antimicrobial compounds.
(FR) La présente invention concerne de façon générale l'élaboration de médicaments, et plus précisément, le criblage de nouveaux composés pour leur activité antimicrobienne. Cette invention concerne des méthodes de production de bibliothèques de peptides mutagénisés exprimés dans des colonies cellulaires. L'association d'un dosage de cellules fluorescentes vivantes/mortes et d'un spectrophotomètre d'imagerie numérique fournit une méthode de criblage en phase solide à haut rendement utilisée pour des colonies ou des bibliothèques synthétiques en micro-réseaux. Ce dosage permet de cribler l'activité antimicrobienne dans au moins 105 à 106 colonies par expérience. Par ailleurs, ces méthodes permettent de produire et de générer des composés antimicrobiens.
