(EN)A method of identifying interactions between polypeptides is provided. The method is effected by: (a) expressing in a cell substantially lacking Ras activity: (i) a first polynucleotide encoding a first polypeptide being capable of interacting with a plasmalemma of the cell; and (ii) a second polynucleotide encoding a second polypeptide fused to a cytoplasmic Ras mutant, the cytoplasmic Ras mutant being capable of the Ras activity if mobilized to the plasmalemma of the cell; and (b) detecting a presence or absence of the Ras activity in the cell, the Ras activity being indicative of a possible interaction between the first polypeptide and the second polypeptide.
(FR)L'invention porte sur un procédé d'identification d'interactions entre des polypeptides. Ce procédé consiste à: (a) exprimer dans une cellule l'activité quasiment nulle de Ras: (i) un premier polynucléotide codant un premier polypeptide capable d'avoir une interaction avec une membrane cytoplasmique de la cellule; et (ii) un second polynucléotide codant un second polypeptide fusionné à un mutant Ras cytoplasmique, ce mutant Ras cytoplasmique étant capable d'activer Ras s'il est immobilisé sur la membrane cytoplasmique de la cellule; et (b) détecter la présence ou l'absence d'activité de Ras dans la cellule, l'activité de Ras indiquant une éventuelle interaction entre le premier et le second polypeptide.
