Abstract:
(EN) Lymphocytes from chronic lymphocytic leukemia (CLL) patients have been found to be readily killed by ultra-violet light-C (UVC) radiation. Cells from healthy donors were minimally affected by doses of UVC 10 times higher than those which caused dramatic drops in the metabolism of CLL cells and eventual death. Irradiated cells from CLL patients and from healthy individuals all demonstrated a number of single strand DNA breaks and alkali-labile sites compared to unirradiated control cells. The extent of DNA damage to both healthy and CCL cells is dose dependent. However, the CLL cells demonstrated more extensive DNA fragmentation and an inability to undergo self-repair. The heightened sensitivity to UVC radiation of lymphocytes from CLL patients is used to provide an excorporeal treatment of CLL lymphocytes followed by the re-introduction of the treated lymphocytes to the patient.
(FR) Selon cette invention, il s'avère possible d'éliminer facilement les lymphocytes prélevés chez des patients atteints de leucémie lymphoïde chronique (CLL) par rayonnement ultraviolet C (UVC). Les cellules provenant de donneurs sains ont été infectées au minimum par des doses d'UVC 10 fois supérieures à celles qui ont provoqué des baisses dramatiques du métabolisme des cellules contaminées par CLL, et éventuellement la mort. Les cellules irradiées prélevées chez des patients atteints de CLL et chez des individus sains présentaient toutes un nombre de ruptures dans un brin d'ADN unique et de sites labiles alcalins comparées aux cellules non irradiées. Le dommage à l'ADN s'étendant aux cellules saines et aux cellules infectées dépend du dosage. Toutefois, les cellules infectées par CLL présentaient une fragmentation d'ADN plus importante et une incapacité à s'autoréparer. La sensibilité accrue au rayonnement UVC des lymphocytes des patients atteints de CLL est utilisée pour effectuer un traitement extracorporel des lymphocytes CLL qui, après traitement, sont réintroduits dans le corps du patient.
