Abstract:
(EN) An expression system useful for the detection and isolation of a polypeptide capable of regulating a transduction pathway, the expression system comprises (a) a first expression construct including a first coding region encoding a reporter molecule, the first coding region being under transcriptional control of a cis acting regulatory sequence element, the cis acting regulatory sequence element being regulatable by a trans acting regulator of the transduction pathway; and (b) an expression library including a plurality of second expression constructs, each of the plurality of second expression constructs of the expression library including a second coding region encoding for one of a plurality of polypeptides, the second coding region being under a transcriptional control of a promoter, such that when the first expression construct and a second expression construct of the plurality of second expression constructs of the expression library are introduced into a cell, the cell endogenously expressing the trans acting regulator of the transduction pathway, a level of expression of the reporter molecule in the cell is indicative of regulation of the transduction pathway by a specific polypeptide of the plurality of polypeptides expressed by the cell from the second expression construct.
(FR) L'invention concerne un système d'expression utile dans la détection et l'isolement d'un polypeptide, capable de réguler une voie de transduction. Le système d'expression comprend (a) une première construction d'expression comprenant une première région codante codant une molécule rapporteuse, la première région codante étant sous le contrôle transcriptionnel d'un élément de séquence régulatrice agissant en cis, ledit élément de séquence régulatrice agissant en cis pouvant être régulé par un régulateur agissant en trans de la voie de transduction, et (b) une banque d'expression comprenant une pluralité de secondes constructions d'expression, chacune desquelles comprend une seconde région codante codant un des polypeptides, cette seconde région codante étant sous le contrôle transcriptionnel d'un promoteur, de façon que lorsque la première construction d'expression et une des secondes constructions d'expression sont introduites dans une cellule, celle-ci exprimant de façon endogène le régulateur agissant en trans de la voie de transduction, un niveau d'expression de la molécule rapporteuse dans la cellule indique la régulation de la voie de transduction par un polypeptide spécifique de la pluralité de polypeptides exprimés par la cellule de la seconde construction d'expression.
