Abstract:
(EN) A gene of an enzyme participating in the synthesis of GDP-L-fucose. More particularly speaking, Arabidopsis-origin GDP-4-keto-6-deoxy-D-mannose-3,5-epimerase-4-reductase gene and a process for producing GDP-L-fucose by using this enzyme. The enzyme encoded by this gene is: (a) a protein containing the amino acid sequence represented by SEQ ID NO:1; or (b) a protein containing an amino acid sequence derived from the amino acid sequence of SEQ ID NO:1 by deletion, substitution, addition or insertion of one or more amino acid residues and having a GDP-4-keto-6-deoxy-D-mannose-3,5-epimerase-4-reductase activity. Thus, GDP-L-fucose, which is essentially required in adding fucose having very important functions to sugar chains, can be efficiently produced in a large amount.
(FR) L'invention porte sur un gène d'enzyme intervenant dans la synthèse de la GDP-L-fucose et en particulier sur le gène de la GDP-4-kéto-6-désoxy-D-manose-3,5-épimérase-4-réductase tiré de l'Arabidopsis et sur un procédé d'obtention de la GDP-L-fucose à l'aide de ladite enzyme. L'enzyme codée par ce gène est: (a) une protéine contenant la séquence d'acides aminés représentée par SEQ ID NO:1; ou une protéine contenant une séquence d'acides aminés dérivant de la SEQ ID NO:1 par suppression, substitution, adjonction ou insertion d'un ou plusieurs groupes d'acides aminés et présentant l'activité de la GDP-4-kéto-6-désoxy-D-manose-3,5-épimérase-4-réductase. On peut ainsi produire efficacement en grande quantité la GDP-L-fucose, essentielle pour ajouter à des chaînes glycoliques les très importantes fonctions de la fucose.
